CD33-directed Antibody-Drug-Conjugates (ADCs) include gemtuzumab ozogamicin and newer auristatin- or PBD-based constructs.

These ADCs are used in combination with anthracycline-based induction and cytarabine (a pyrimidine analogue) backbones for CD33⁺ AML treatment.

More rational ADC scheduling is being considered due to toxicity and therapeutic-index constraints.
