The cGAS-STING pathway is a crucial part of the immune system. It recognizes double-stranded DNA, various endogenous DNA species as byproducts that arise in different contexts of cellular stress (e.g., cytosolic DNA) and translates this into transcriptional and other cellular effector responses such as triggering an immune response. Its canonical role is in antiviral defense. In this function, cGAS-STING influences tissue homeostasis and antitumor immunity, but is also associated with a range of inflammatory diseases.
