Peptide-Based Antimicrobials and Siderophore Conjugates are two classes of potential next-generation treatments for MDR pathogens.

  • Can selectively kill bacteria.
  
  
  • Efficacy is context-dependent.
  
  
  • Challenges include host-driven proteolysis, poor bioavailability, and limited tissue penetration.
  
  
  
  


• Siderophore Conjugates:
  
  
  • Use iron uptake systems for targeted delivery.
  
  
  • Depend on bacterial receptors that may be variably expressed, suppressed, or lost during infection.
  
  
  • Example: Cefiderocol's failure in certain clinical settings shows the vulnerability of these approaches.
