Enhancer of Zeste Homolog 2 (EZH2) Histone-Lysine N-Methyltransferases is a validated therapeutic target in follicular lymphoma because gain-of-function EZH2 lesions are common and drive abnormal H3K27 trimethylation and germinal-center persistence. In FL, the clinically established EZH2 inhibitor is tazemetostat, with the strongest activity seen in EZH2-mutant relapsed/refractory disease.
